Táto diplomová práca sa zameriava na vývoj HPLC metódy pre stanovenie vybraných bioaktívnych látok z rastliných extraktov. Presnejšie sa práca zaoberá separáciou vybraných flavonoidov a to rutínu, hesperidínu, naringínu, fenolpropanoidov: dihydrokumarínu, eskulínu a escínu a derivátom antrachinónu hypericínu. Tieto látky pochádzajú z rastlín: Pagaštan konský (Aesculus hippocastanum), Pohánka tatárska (Fagopyrum tataricum (L.)), Komunica lekárska (Melilotus officinalis (L.)), Ľubovník bodkovaný (Hypericum perforatum) a Grep (Citrus paradisi). Pre toto stanovenie boli vyvinuté tri metódy. Metóda s gradientom bola použitá pre stanovenie rutínu, hesperidínu, naringínu, dihydrokumarínu a eskulínu. Escín a hypericín boli potom analyzované pomocou izokratických metód. |

This diploma thesis aims at the development of the HPLC method for identification and quntification of chosen bioactive substances from herbal extracts. More specifically, the thesis deals with the separation of particular flavonoids (rutin, hesperidin, naringin), phenylpropanoids (dihydrocoumarin, aesculin, aescin) and anthraquinone derivatives (hypericin). These substances originate from the following herbs: European horse-chestnut (Aesculus hippocastanum), tartary buckwheat (Fagopyrum tataricum (L.)), yellow sweet clover (Melilotus officinalis (L.)), St John's wort (Hypericum perforatum) and grapefruit (Citrus paradisi). For the above-mentioned determination, three methods were developed. The gradient method was used in the identification and quantification of rutin, hesperidin, naringin, dihydrocoumarin and aesculin. Aescin and hypericin were then analyzed by means of isocratic methods. |
